VI.2 Examination of Decaying Leaf Tissue

Aim

To observe structural changes in plant tissue during decay.

Specimen & Apparatus

  • Decaying leaf material
  • Forceps or needle
  • Slide and coverslip
  • Water
  • Compound microscope

Method

  1. Select a small portion of softened leaf tissue.
  2. Place on a slide with a drop of water.
  3. Gently tease apart the material if required.
  4. Apply a coverslip carefully.
  5. Examine under low and then higher magnification.

Observations to Make

  • Breakdown of cell structure.
  • Presence of microorganisms or fungal growth.
  • Changes in colour or texture of tissue.
  • Differences between intact and degraded areas.
