I.1 Examination of the Epidermis of a Leaf

Aim

To observe the arrangement and form of epidermal cells in a leaf.

Specimen & Apparatus

  • Fresh leaf (e.g. ivy, privet, or laurel)
  • Forceps or needle
  • Slide and coverslip
  • Water
  • Compound microscope

Method

  1. Remove a small portion of the leaf.
  2. Gently peel a thin strip of the epidermis from the surface using forceps.
  3. Place the peel in a drop of water on a slide.
  4. Apply a coverslip carefully to avoid air bubbles.
  5. Examine under low power, then increase magnification.

Observations to Make

  • Shape and arrangement of epidermal cells.
  • Appearance of cell walls.
  • Presence or absence of chloroplasts.
  • Regularity or variation in the cellular pattern.
