I.1 Examination of the Epidermis of a Leaf
Aim
To observe the arrangement and form of epidermal cells in a leaf.
Specimen & Apparatus
- Fresh leaf (e.g. ivy, privet, or laurel)
- Forceps or needle
- Slide and coverslip
- Water
- Compound microscope
Method
- Remove a small portion of the leaf.
- Gently peel a thin strip of the epidermis from the surface using forceps.
- Place the peel in a drop of water on a slide.
- Apply a coverslip carefully to avoid air bubbles.
- Examine under low power, then increase magnification.
Observations to Make
- Shape and arrangement of epidermal cells.
- Appearance of cell walls.
- Presence or absence of chloroplasts.
- Regularity or variation in the cellular pattern.
